The Pharos is the name of the lighthouse. The answer's not the lighthouse, but it is something of the same scale in history, a greater claim to fame than the lighthouse.
I hadn't thought about the canal. It's a fairly big deal, but not the biggest of the time, which was, if I recall correctly (I haven't looked it up so I may be wrong), connecting the Red Sea and the Nile. The Egyptians had been making canals of different sizes for thousands of years by that point. But it's also a fairly big deal in Alexandria because of location. Good answer, but still not what I'm thinking of.
The original part of this was completed under Ptolemy Soter, and the idea was possibly started by Alexander himself, though the building of what we call it today did not begin until Ptolemy II Philadelphus. Once the addition was begun, it never ended until it was destroyed in stages, by the Romans first and then later "daughter" or "sister" versions by the Christians in the 4th century.
To complete this great quest, laws were passed that any traveller to the city would be searched and a certain class of items would be seized, with replicas of the seized property returned to owner at a later date.

We have a winner!
The Musaion was built to be a center of learning and knowledge. It was completed under Ptolemy I Soter with the intent of becoming the greatest university (as we would think of the term) in the known world. This was not the revolutionary idea, though, as previous schools had also tried for this goal.
The revolutionary idea was that the Ptolemies tried to get a copy of every scroll or book ever created, and not only that, they wanted those books to be used for teaching, not just stuffed away in a shelf and forgotten. To that end, any traveller who entered the city was searched, as was their caravan/ship, and then a copy of the book was made and the copy returned to its owner within a few days. Can you imagine? Every scroll ever created in any language ever anywhere (well, as close as they could get anyway). This made Alexandria the greatest center of learning that has ever existed on this planet, even compared with universities today.

The Royal Library is kind of my obsession. It was my specialization in Classical Civ in college. Actually, Arsinoe II is kind of my obsession, but it's all wrapped up together. So I don't have a singular source for my question. There are a few books out on the subject though and I can offer recommendations if anyone wants them.
If I'd had the money to finish college I'd be in Alexandria attached to an archeological investigation of some kind. I really want to be out there looking for the location of the temple Ptolemy II Philadelphus built for Arsinoe II when she died and he had her deified. One written account says her cult statue was made of yellow topaz (or similar stone) and floated through use of magnets. I know it's not likely to be literally true, but it's a pretty fascinating idea. I thought about saving that for a question, but the written account is pretty obscure if you don't read greek
